Abstract

The composition of the alpha-amanitin-resistant RNA polymerase induced in baculovirus-infected cells will be determined. A combination of approaches are envisaged. These include direct sequencing of isolated polymerase subunits and the use of polyclonal antibodies raised to synthetic peptide sequences to identify putative polymerase components. A further novel approach will involve the addition of epitope coding sequences to putative virus polymerase genes, which can be detected using monoclonal antibodies. Evolutionary relationships of the baculovirus RNA polymerase with other enzymes will also be determined.
