Abstract

Through an understanding of the 3-D structures of viral proteins and of their packing in viral particles, we can begin to consider strategies with which to interrupt virus propagation in infected plants. Our objective is to design novel antiviral proteins for expression in transgenic rice plants based on the crystal structures of proteins involved in Rice Dwarf Virus transmission and replication. To this end, funding for a research assistant at the RA1B level and limited equipment is requested who will perform initial purification, crystallisation and X-ray characterisation of a number of viral proteins. Full structure solution of a capsid protein may be possible within the grant period but it is intended that subsequent proposals will build on this work to solve individual structures and pursue antiviral design studies.
