Abstract

A wide range of stimulus-response pathways in plants are mediated through elevation of cytosolic free Ca2+. At least four separate classes of voltage- and ligand-gated Ca2+ release channels reside at the vacuolar membrane, but their roles in mobilising Ca2+ in response to specific stimuli is unknown. Channel-specific agonists (including cyclic ADP-ribose) and antagonists will be micro-injected into guard cells subjected to stimuli known to elevate Ca2+ and the characteristics of any Ca2+ transients will be monitored. The work will extend to testing the hypothesis that interactions between channel types is involved in regenerative Ca2+ signalling and hence in the generation of stimulus-specific Ca2+ signals.
