Abstract

The proposed research will determine whether Spo0A controls the onset of solventogenesis in saccharolytic clostridia by a direct or an indirect mechanism. The binding of B. subtilis and C. beijerinckii Spo0A to 0A boxes located in the upstream regions of clostridial genes whose expression is modulated at the onset of solventogenesis will be tested by gel retardation and footprinting experiments. This approach will be complemented by studying the expression of sol-lacZ gene fusions in B. subtilis in different genetic backgrounds. A possible role for AbrB (itself under spo0A regulation) in control of solventogenesis will also be explored. Modified forms of C. beijerinckii sol genes created in vitro and tested in B. subtilis will be used to replace the wild type alleles in the bacterial chromosome to test whether the deduced control mechanisms operate in vivo. The role of DNA elements that promote degeneration of solventogenesis in the wild type strain will be elucidated.
