Abstract
A chemotaxis operon identified in Rhodobacter sphaeroides and related species has major differences in both gene arrangement and content compared to enteric bacteria: (i) two cheY genes (ii) no identified cheZ. This, coupled with the observed behaviour of these species, suggests that the alpha-subgroup of proteobacteria may use a different chemotactic system to species in the gamma -subgroup. We will (a) Measure the phosphorylation kinetics of CheY- 1 and CheY-2 by Che-A to identify whether both CheYs are substrates for Che-A. (b) Identify the chemosensory pathway operating through the 2 Che-Ys by both mutagenesis and the kinetics of responses to the photorelease of caged compounds. (c) Use a fluorescent- MCP fusion protein to identify whether the MCP(s) is targeted to the pole or flagellum.
