Abstract
1) Cloning, sequencing and expression of citrate synthase genes from Archaea that span the phenotypic range of temperature and halophilicity. 2) Analysis of the 3D-structure of these enzymes by X-ray crystallography, molecular modelling and molecular dynamics simulation. 3) Prediction of structural features conferring extreme stability and testing of these predictions by site-specific mutagenesis.
