Abstract
The project will (a) clone the genes encoding the pathway for 4- nitrotoluene catabolism in two Pseudomonas strains, (b) obtain a physical map of their organisation (c) express the genes in suitable plasmid vectors both to elucidate some of the later steps in the pathway and to obtain high levels of enzyme activities to study the enzymology of individual steps, (d) draw conclusions as to the evolutionary relationship with analogous steps in the catabolism of alkyl toluenes.
