Abstract

We have recently purified a tobacco protein (3' TNS RNA binding protein) that binds to the 3'-terminus of the negative-strand RNA of cucumber mosaic virus (CMV). A cDNA clone of the 3' TNSBP will be obtained, sequenced and expressed in bacterial and baculovirus systems. The RNA binding sites on the negative-strand of CMV RNAs 1, 2 and 3 will be defined. The ability of the 3'-TNS binding protein to interact with other proteins of the CMV replicase complex will be investigated. The effect of the 3'-TNS RNA binding protein on the production of positive and negative RNA strands by the CMV replicase in vitro will be determined.
