Abstract
We will localise primary carbohydrate metabolism in source leaves of barley to the level of the cell, especially epidermis, mesophyll and parenchymatous bundle sheath. We will ask: do vacuolar sucrose and vacuolar acid invertase accumulate in the same cell type? Is sucrose (and fructan) stored preferentially in non-photosynthetic cells, especially when photosynthesis exceeds demand? Is fructan stored in cells with high vacuolar sucrose? We will use single-cell sampling with the pressure probe, antibodies and immunogold labelling, and C-14 techniques, on both control leaves and those induced to accumulate carbohydrate.
