Abstract
A recently-identified Brassica napus cutinase, responsible for the penetration of stigmas by pollen tubes, will be investigated at the level of the polypeptide and the gene. Immunogold electron microscopy will be used to monitor the accumulation of the enzyme during grain development, its secretion into the intine layer of the wall and emission at the pollen tube tip during stigmatic penetration. Newly-acquired biochemical and molecular data will be used in the identification and cloning of the cutinase gene; in parallel PCR-based approaches we shall (a) extend promising work using primers based on fungal cutinase sequence data, and (b) use currently-available N- terminal sequence from the pollen cutinase itself to construct primers for amplification. Northern and in situ hybridisations will establish the timing and site of transcript accumulation, and study of genomic clones will reveal both the structure of the gene and identify upstream regulatory sequences directing pollen-specific expression.
