Abstract
This study sets out to investigate the effects of changing the levels of the photosynthetic carbon reduction (Calvin) cycle enzymes fructose- 1, 6-bisphosphatase (FBPase) and sedoheptulose-1, 7-disphosphatase (SBPase) on carbon dioxide fixation and biosynthesis of starch and sucrose. Mutant lines will be obtained by transforming tobacco plants with sense and antisense constructs prepared using cDNA clones encoding chloroplast FBPase and SBPase. The transgenic plants produced will be analysed for changes in protein levels by western blotting and enzyme assay techniques. Selected lines with altered levels of FBPase or SBPase will be examined biochemically and physiologically. Non-invasive techniques will be used to assess their photosynthetic performance in a range of environmental conditions and will allow us to calculate flux control coefficients. The impact of altered FBPase and SBPase activities on partitioning between starch and sucrose will be analysed in the transgenic plants.
