Abstract
We have recently isolated an RNA-dependent RNA polymerase from red clover necrotic mosaic-virus (RCNMV)-infected plants and shown it to contain two virus-encoded and two plant-encoded proteins. The genomic and subgenomic promoter elements in the (+) and (-) strands of the virus RNA will be defined by in vitro mutagenesis of RNA templates in conjunction with RNA binding and polymerase assays. The proteins in the virus RNA-replicase complex that bind to these promoter regions will be identified by u.v. cross-linking and the protein binding domains will be located by deletion, alanine-scanning and site-directed mutagenesis.
