Abstract
Receptor and G-protein activated phospholipase C-beta isoforms messengers, inositol trisphosphate and diacylglycerol. Recombinant PLCbetas will be used to raise panels of monoclonal antibodies which will be selected for interaction at G-protein, substrate and interfacial recognition sites. Epitope mapping approaches will reveal the spatial organisation and sequence relationships of these sites. In addition, these immunological tools will be used in subcellular localisation and enzyme mechanistic studies. These studies are an essential prelude to any attempt at rational design of potent inhibitors of this important group of enzymes.
