Abstract
From a chance observation, we wish to make nucleoside and nucleotide prodrugs from 5- fluorouracil and 4-thio-2-deoxyribose. The following points will be addressed : the conditions necessary for stereoselective synthesis; the synthesis of novel prodrugs of the 5'- nucleotide which are labile to chemical hydrolysis, enzymatic attack or both; the synthesis of such prodrugs carrying targeting molecules for delivery to specific locations and investigating the cellular metabolism and mode of action of these analogues. This information will then be used to feed back into the synthesis to enhance further the selectivity of these compounds.
