Abstract

A strategy is proposed to develop a quantitative framework for defining the pharmacokinetics of oligonucleotides in vivo. Using specific assays, a physiologically-based approach will be taken to characterise and model the pharmacokinetics of oligonucleotides in plasma and tissues after single and multiple administration to mice and rats. Detailed kinetic aspects of hepatic uptake and distribution will be explored in the isolated perfused liver.
