Abstract

We propose to generate strains and mutants of yeast to investigate the controls of mevalonate metabolism at branch-points leading to the formation of squalene, ubiquinone, carotenoids, dolichols or prenylating compounds. To this end, we shall generate sterol auxotrophs of yeast which are deficient in squalene epoxidase or other enzymes of isopentenoid metabolic pathways, and which possess elevated levels of hydroxymethyl glutaryl CoA reductase activity.
