Abstract
The main aim is a joint mathematical and biological study of the spatial dynamics and gene regulation of a microbial ecosystem. Bacterial Green Fluorescent Protein reporters will be used with a continuous flow system consisting of Salmonella typhimurium and Acanthamoeba polyphaga. Measurement will be conducted by direct microscopy, with parameters of the system and probability distributions of the microscopic events ascertained by analysis of video images. The salmonella-amoeba interaction will be clarified, with particular reference to changes with environmental conditions such as temperature and the degree to which the interaction influences the expression of virulence determinants. Stochastic mathematical models of individual cell gene regulation will be used in the construction of spatial population models and statistical techniques to compare the data (video images) with model predictions. The models will elucidate the control of genes at both the individual and population levels in response to environmental signals. (Joint with grant MMI09727)
