Abstract

This application is a request for funds to set up a high resolution, high sensitivity, digital video, ratio-imaging workplace in the Department of Anatomy and Physiology at the University of Dundee not yet available here. The system should allow the 3-dimensional tracking of fluorescently labelled cells during embryogenesis. Cell-cell signalling and signal transduction processes will be followed by fluorescent ratiometric measurement of changes in ion concentrations (Ca2+ and H+) associated with particular signal transduction events as well as by direct measurement of dynamical changes in protein-protein interactions by fluorescence resonance energy transfer (FRET) between blue and green fluorescing fusion proteins formed with multimeric signal transduction components.
