Abstract
A confocal microscope facility is requested for use in a number of projects in which green fluorescent protein (GFP), or its spectral variants will be employed to investigate: (1) development in roots and in leaves; (2) modification of Na+ transport activities in specific cell types; (3) subcellular targeting of proteins to the nucleus and chloroplast; and (4) the dynamics of the vacuolar compartment in stomatal guard cells. Several of the projects will use unique Arabidopsis lines that have cell-specific expression of a GAL4-GFP construct that allows cells expressing GAL4 to be identified from their bright fluorescence. These lines will be crossed with others containing constructs with toxin or Na+ transporter genes controlled by a GAL4-dependent promoter. This will activate these genes in the cells expressing the GAL4-GFP construct. Expression of the toxin will cause genetic ablation of the cells allowing their role in developmental processing to be assessed. Expression of the Na+ transporters will cause changes in the Na+ transport capacity of the targeted cells with possible enhancement of salt tolerance. The chloroplast and vacuole projects will use GFP or one of its variants fused to other proteins to follow the biogenesis and dynamics of these organelles.
