Abstract

The protein structure group at Exeter is formed from a multi- disciplinary team who are involved with the cloning, over- expression, protein characterisation and x-ray structural determination of a number of proteins of importance on biotransformation reactions and with medical relevance. In the last 2 years the group has solved 6 structures with resolution ranging from 1.1 to 2.0 angstrom. Four of the structures were solved by heavy atom derivative isomorphous replacement methods. This proposal is to enhance the x-ray facilities already available in Exeter to make data collection from new crystals of human perixiredoxin, human glucokinase, and of novel enzymes important for biotransformation reactors such as Baeyer-Villiger monooxygenases both type I and type II from Pseudomonas putida and Acinetobacter species, amidohydrolases with opposite stereospecificity for gamma lactam rings obtained from Comomonas and Aurebacterium species, hyperthermophilic novel proteases from the hyperthermophilic Archaeon Thermococcus species. Other projects currently underway are listed in the proposal and include novel esterases, penicillin recognising proteins from Agrobacterial and Fungal sources, tryptophanase, vanadium bromoperoxidase, liver fructose bisphosphate aldolase, thermophilic archaeal glyceraldehyde 3 phosphate dehydrogenase, phosphoglycerate kinase and Thermotoga phosphoglycerat/triose phosphate isomerase natural fusion protein.
