Abstract

Over the last 8 years IAH has developed a bovine immunology programme involving the characterisation of cells and molecules of the immune system considered to be essential to provide a fundamental level of understanding of immune processes, focussing on antigen-presenting cells (APC) and their role in regulating the induction of different types of T cell responses. Advances in this area provide the opportunity to initiate a new programme of work on bovine tuberculosis caused by Mycobacterium bovis, which replicates in APC and is controlled by T cell-mediated immune responses. The investigations will determine how infection of APC affects their ability to stimulate T cells, expression of surface molecules and synthesis of cytokines. The role of T cell populations will be investigated in vivo, and in vitro correlates with immunity established. Bovine tuberculosis has been targeted because of its increasing importance as a cattle disease, with attendant risk to human health, and its relevance as a model for human tuberculosis. Since M. bovis is a human pathogen, many of these studies must be carried out in category 3 containment facilities. This proposal seeks funds for equipment to perform flow cytometric analyses and sorting of bovine cells and within the category 3 facilities.
