Abstract
Enteric infection is a major cause of morbidity and mortality in both human and veterinary species. In addition there is a growing appreciation of the social and economic cost of the effects of allergic reactions to ingested antigens. An understanding of the mechanisms of antigen handling at mucosal surfaces is essential to the effective control of these enteric infections and allergic reactions. This project aims to further characterise both the lamina propria T cell and antigen presenting cell (APC) populations. Previously we have shown that intestinal T cells are a highly differentiated population which are susceptible to apoptosis following activation. We have postulated that clonal deletion of these cells may be one mechanism for the maintenance of oral tolerance and we propose to determine whether activation by locally derived MHC class II+ cells results in cell death in an in vitro model using purified CD4+ T cells and sorted allogeneic dendritic cells. The system will be extended by the inclusion of stomal cell types and cytokines to model the effect of the intestinal microenvironment on T cell activation. The effect of local viral and bacterial infection upon these cell types in vivo will be determined.
