Abstract

Flow cytometry is a powerful technique for the analysis and sorting of cells and particles, based on size, light scattering or fluorescence characteristics. It finds wide applications across the biological sciences, and is indispensable in several types of study. However, appropriate instruments are not widely available in the academic community. This application is for the establishment of a Flow Cytometry facility in the Institute of Biotechnology. The purchase of a Parted PAS-III/PPCS flow cytometer and cell sorter will support a range of projects in the Institute, and allow training of PhD students in this important technology. The instrument has been selected for its versatility, since it will be required in projects across several disciplines, including analysis of cell cycle and growth in plants and plant cultures, studies on the inheritance of DNA molecules in yeast, isolation and analysis of microbial cultures for degradation and biotransformations of morphine alkaloids and explosives, and the use of beaded materials for affinity purification and chemical combinatorial library screening. The provision of this instrument will therefore support both the research and training environments of the Institute of Biotechnology, as well as being made available to interested researchers in other relevant departments.
