Abstract

The objectives of the proposed research programme are to: 1) Determine what regulates the kinetochore localisation and kinase activity of the Bub 1 protein kinase. 2) Determine how cells exit from mitosis in the presence of a damaged spindle and how this induces apoptosis. 3) Test whether the Bub 1 dependant checkpoint pathway plays a role in regulating meiotic chromosome segregation. The proposal covers 3 areas that are linked by either experimental approach or reagents. Each area should provide excellent opportunities for a high quality research programme. Therefore, during the first 2 years initial experiments will be performed in all 3 areas. After 2 years, a decision will be made regarding future directions. If all the preliminary experiments have gone well, then it should be possible to continue in all areas and achieve the stated goals by the end of the fellowship. If only some of the areas prove to be tractable, these will be focused on for the remainder of the fellowship. In addition alternative strategies to address the less tractable issues will be initiated. The remaining three years should provide sufficient time to make significant progress with alternative strategies.
