Abstract

This proposal is to support computing for the structural biological chemistry group at Exeter University headed by Dr J. Littlechild. The computing power available to us at present funded by BBSRC and Wellcome Trust is not of sufficient size to allow us to progress with refinement of high resolution structures of enzymes important in biotransformation reactions or to carry out molecular modelling and dynamics on enzyme/substrate/inhibitor complexes. The Origin 200 computer system in this bid will allow us to work efficiently 'in house' and allow development of our group in this important area of research. Funding for our research comes from project grants supported by both ROPA awards and as part of the Chemical and Pharmaceutical Directorate of the BBSRC and charities such as the Wellcome Trust. We have LINK programme grants with Chiroscience and CASE studentships with SmithKline Beecham, Chiroscience and British Biotechnology. Structural information at high or atomic resolution of enzymes used in chemical synthesis is of vital importance to the pharmaceutical industry for the synthesis of homochiral drugs or synthons. An understanding of the enzyme mechanism which is of both generic and industrial interest will allow redesign of these enzymes to optimise their use in biocatalysis. Support for this equipment bid will help us to achieve these ends.
