Abstract
This application is for funds to purchase a sensitive matrix- assisted laser desorption/ionisation (MALDI) mass spectrometer for MS and MS/MS studies of carbohydrates. The instrument will be based in the Glycobiology Institute and will support several projects including glycosylation studies of molecules involved in the immune response, such as CD-5, CD-48 and CD-59 (both with and without an anchor), the chaperone calreticulin involved in protein folding, antigenic peptides involved in allergic reactions and on the glycosylation of the hepatitis-B virus. Glycolipids from patients suffering from glycolipid storage diseases will be studied with the aim of developing MS-based identification and quantification techniques and for understanding the underlying biochemistry. The instrument will also be used to study the biochemistry and diseases of the central nervous system including the molecules Na/K-ATPase which shows organ-specific glycosylation and the ependymins involved in memory formation. Degenerative diseases such as Alzheimer's disease and CJD will be studied using a proteomics-based approach. A high-throughput system based on enzymatic release of glycans followed by MALDI analysis is being implemented for these projects. The instrument will also be used to develop high sensitivity methods for glycan analysis with particular emphasis on new approaches to the analysis of anionic and O-linked sugars.
