Abstract
This proposal is for a transgenic controlled environment plant growth facility at IACR- Long Ashton Research Station that will be used to study the function and to evaluate the commercial potential of plant genes that we have cloned. These include (i) a gibberellin 20-oxidase that we have shown to have the potential to alter endogenous GA-levels in plants and thereby affect elongation growth and flowering, (ii) a GPCR homologue from Arabidopsis that we have shown effects sensitivity to cytokinins and influences time of flowering and apical dominance, (iii) the Arabidopsis developmental regulator ABI 3 which is required to induce embryo dormancy and has potential for manipulation of preharvest sprouting in wheat, (iv) antifungal proteins that may have potential as novel control agents for Septoria leaf spot diseases of wheat (v) TIP1, a gene that controls differentiation in pollen tubes and root hairs. Experiments proposed with these genes will involve the use of transgenic plants capable of transferring novel genetic material to UK plant species. The pathology programme will involve the use of genetically manipulated pathogens. Thus, containment level B conditions are required. Furthermore, because we will study the impact of these genes on developmental events during flowering and seed development and their regulation by environmental factors particularly light quality, photoperiod and temperature it is essential that controlled environment rooms are used.
