Abstract
This proposal is for research aimed at investigating the dynamic events occurring during the transduction of environmental signals in plants. Calcium signalling will be imaged in intact whole plants responding to different abiotic stresses, by the use of the calcium- activated photoprotein, aequorin. Aequorin will be expressed in transgenic Arabidopsis thaliana. Using a highly-sensitive photon-counting camera (for which the funds are requested) we will be able to measure both spatially, and in real time, calcium signalling in intact whole plants responding to cold, heat, oxidative stress, drought and mechanical stimulation. In this way we will be able to 'map' the contributions of this important signal transduction component, in different tissues and cells. This will allow for the first time an understanding of how the information from these different stresses, received by plants, are then converted into a specific response and also what the commonalities/interactions between these pathways are at the level of calcium signalling. Additionally we will use this imaging approach to screen for calcium signalling mutants of Arabidopsis. This will allow for the first time the identificaton of specific mutations early in abiotic stress transduction cascades.
