Abstract

The applicants have established a collaborative research agreement with Amersham International PLC to develop new, intensified high throughput screening assays and procedures that extend the use of the company's Cytostar and SPA technologies. The Cytostar-T and SPA technologies permit the real-time analysis of a wide range of cellular events by optical imaging of radioisotope uptake by surface cultured cells in 96-well scintillating microplates and on scintillating microspheres (respectively). These proprietary technologies have been adopted by the majority of leading pharmaceutical companies and our collaboration involves research on new drug and metabolite uptake assays, vector targeting and reporter gene assays, development of the SPA assay and the use of this assay for screening combinatorial peptide libraries. To meet the demand for yet higher throughput screening, metabolism and toxicology assays, and to permit the development of more convenient fluorophore based assays, it will be necessary to extend the range of optical imaging to much lower light levels than are currently monitored. We therefore seek to exploit the imaging of ultra-low light levels using digital CCD camera technology and request funding for the Tundra ultra-low light imaging system to conduct high resolution and dynamic detection of fluorescence and luminescence in scintillating microplates.
