Abstract
To provide the John Innes Centre, with a unique, anaerobic, multi- mixing, stopped-flow Fourier transform infra-red spectrometer (SF- FTIR) for transient kinetic studies, principally of small molecule binding and activation of metal sites in enzymes and chemical complexes, with the aim of elucidating mechanisms of catalysis at atomic resolution. Nitrogenase and related chemistry; Oxalate oxidase germin; Cu-containing nitrite and N20 reductases and related chemistry; Cyt. bo3, a haem-Cu oxide (with UEA); Cyt.cd1 nitrite reductase (with Oxford) will be studied initially. These are Foresight priorities in catalysis, chemical nitrogen fixation and environment/pollution, denitrification and water purification. The equipment is a development of a prototype instrument built by Dr C. Wharton (Birmingham, Biochemistry) for proteinase and protein folding studies. The feasibility of using S-F FTIR for metallo-protein studies and related chemistry has recently been shown by the applicant at Birmingham (George et al, J. Amer. Chem. Soc. 1997, 119, 6450-6451). The JIC facility will be made available to other groups on the Norwich Research Park, principally members of the BBSRC Centre for Metallo-protein Spectroscopy and Biology at UEA (Directors Prof. A. J. Thomson & Prof. C. Greenwood) and the UK research community in academia (e.g. Prof. S. J. Ferguson, Oxford, Biochemistry) and industry (e.g. Unilever). In addition to Unilever, a financial contribution has been obtained from Hi-Tech Ltd, a UK SME who will develop and supply a new multi- mixing drive and sample handling unit.
