Abstract
The laboratories interested in skeletal research at UCL form the largest such grouping in the UK, with longstanding collaborations. We need an efficient method for detecting changes in skeletal phenotype in the many strains of transgenic mice that we now study. The method of choice is the GE/Lunar PIXImus dual energy X-ray absorptiometer, a high throughput instrument specifically designed for measuring bone mineral density in small, sedated rodents. The instrument would be used monitoring bone density in mice deficient in: P2 receptor or ecto- nucleotide pyrophosphatase; connexin-43-deficient mice; tartrate-resistant and lysosomal acid phosphatases; cathepsin K; Cbfa 1; the glutamate transporter GLAST; biglycan and decorin; collagen type X; interleukin-4 and interferon-gamma. Additional studies with PIXImus will include: bacterial bone pathology and the osteolytic role of molecular chaperones; osteopetrosis; the osteolytic action of acidosis and hypoxia; antiresorptive drug development. In view of the concentration of the molecular biology/transgenic mouse research at UCL, there are likely to be many unforeseen future applications. The proposal represents excellent value for money and will provide a new focus of collaboration for our laboratories.
