Abstract
This is a multi-user grant application to provide a state of the art mass spectrometer to underpin biological and biochemical research in the Chemistry, Biochemistry, and Pathology Departments and at the Oxford Centre for Molecular Sciences (OCMS). We request 58 percent funding for a Micromass Q-Tof micro machine which is a bench-top Quadrupole/orthogonal acceleration time-of flight instrument capable of on-line HPLC-MS and MS-MS analyses. The remainder of the funding comes from Micromass (with whom we have a long-standing collaboration), the Chemistry Department and OCMS. In addition to routine mass analyses the machine will be used for the study of non-covalent protein-protein interactions, and post-translational modifications. Proteomic studies will benefit from the ability of the instrument to perform simultaneous MS and MS-MS analyses on HPLC-MS analyses, providing both molecular weight and fragmentation/sequence (structural) data. The upper m/z range of 20,000 will allow detection of non-covalent, protein-substrate and protein-protein complexes. A wide range of research interests will be supported: analysis of the sub-nuclear structure, polymerases, modular proteins, glycoprotein chemistry, bespoke oligomers/peptidomimetics, cell cycle regulatory proteins, immunochemistry, enzymology and metabolism, and peptide sequencing.
