Abstract
Mass spectrometry is rapidly becoming a de facto standard tool in protein laboratories. We aim to install a mass spectrometer suitable for analysis of liquid phase samples, and over an extended mass range, in the Faculty of Veterinary Science at Liverpool. The specification lends itself to multiple projects that will use the machine with quick turnaround, and permits rapid examination of a single sample, interspersed with longer term and sequential analyses. The system will be used to generate metabolite, protein and peptide mass data from a range of projects, including skeletal muscle and yeast proteomics, in the molecular recognition aspects of limited proteolysis, phenotypic characterisation of individual molecules, the identification of intracellular binding cognates of matrix metalloendopeptidases, the analysis of disease indicators and assessment of toxins. This application is part of an integrated plan of development of mass spectrometric capabilities between Liverpool and UMIST, wherein simple MS analyses at Liverpool will provide basic data to inform subsequent experiments or to identify those samples that require more sophisticated mass spectrometry analysis such as tandem MS using the facilities contained within the Michael Barber Centre for Mass Spectrometry at UMIST.
