Abstract

Funding for state-of-the-art protein purification equipment is requested to underpin the work of several research groups in Bristol. This includes structural and mechanistic studies on polyketide synthases from fungal and bacterial sources and the enzymes involved in post-polyketide modifications, with a view to producing new bioactive molecules; studies on the enzymes of amino acid and other primary metabolic pathways aimed at the design of mechanism-based inhibitors; structural and mechanistic studies on individually expressed domains of fatty acid synthases and the novel enzymes involved in fatty acid elongation processes; isolation and characterisation of chlorinating and other enzymes from marine metabolic pathways to study their potential for biotransformation. These studies involve extensive collaborations within Bristol and with several groups in the UK and elsewhere.
