Abstract

Request is made for a contribution towards the purchase of a novel CD-spectrometer capable of both steady-state and time-resolved measurements. The instrument will be used to support a number of ongoing projects in Bacterial Physiology and Biochemistry Group based in the School of Biological Sciences at UEA. These projects encompass three main themes: assessing protein conformation, protein:protein interactions and redox-linked conformational changes. The principal requirement is for far-UV CD spectroscopy but there is also a need to use CD to examine the environment of some co- factors such as flavins. A particular feature of this instrument is the ability to make absorbance and CD measurements on the same sample. This is essential for a number of projects where we seek to temporarily relate changes in the redox- or ligation-state of a chromophore to conformational change. In addition we have clearly identified a wider user community at UEA and on the nearby Norwich Research Park who will be able to use this instrument to support work in the areas of metalloproteinase biochemistry, plant disease resistance and structural biology.
