Abstract

Investment is urgently needed for equipment to support the following investigations into gene function in antibiotic-producing Streptomyces: 1). systematic transposon mutagenesis of the model species, Streptomyces coelicolor, relating phenotype to genotype in each mutant; 2). molecular characterisation of the milbemycin polyketide synthase gene cluster; 3). molecular genetic characterisation of the control of gene expression in response to changes in osmolarity; 4). elucidation of the control of gene expression in Streptomyces by plant signalling compounds; 5). analysis of a novel type of site-specific DNA modification; and 6). investigations into the genetics of oxidative stress and cell ageing in antibiotic-producing mycelia. The areas of research also provide an excellent training ground for postgraduate students.
