Abstract
Gibberellins are endogenous growth regulators that are involved in all aspects of plant development. The GA-biosynthetic pathway is regulated by a range of endogenous and environmental factors including light, temperature and feedback/feedforward by bioactive GAs. We propose to use the promoter from a feedback-regulated GA-biosynthetic gene coupled to the firefly luciferase reporter gene to study the response of this promoter to different chemical and environmental stimuli, in real time. We also propose to identify components of the regulatory mechanisms by identifying mutants that affect expression of the reporter gene under a range of different experimental treatments. To detect the low level of light emitted by luciferase we require a highly-sensitive, liquid nitrogen- cooled CCD camera and image analysis software that will allow us to quantify changes in luciferase activity and to screen large numbers of mutant seedlings to identify individuals with altered responses.
