Abstract

A flow cytometry analyser and confocal laser scanning microscope are required to support a programme of work investigating the pathogenesis and pathology of transmissible spongiform encephalopathies (TSEs) in experimental models. The objectives of this programme are to identify the cells and mechanisms involved in: (1) The spread of infection from the site of entry (e.g. skin, gut) to sites of replication in lymphoid tissues; (2) Replication of infectivity within lymphoid tissues; (3) Spread of infection to and within the CNS; and (4) Neurodegeneration and loss of CNS function. The programme includes studies of the effects of manipulations of the immune system on pathogenesis in lymphoid tissues, as well as detailed morphological analyses of pathological changes in both lymphoid and nervous systems. An understanding of the pathogenesis of TSEs is necessary for the design of therapeutic or prophylactic strategies and will also aid risk assessment in relation to accidental exposure to potentially contaminated tissues.
