Abstract

Molecular diagnostics is moving from low throughput, heterogeneous and mostly manual technologies to higher throughput, closed-tube and automated methods. A highly favoured signalling technology for such assays is fluorescence and a number of techniques relying upon energy transfer between a fluorophore and a proximal quencher molecule have been described. In these methods, dual labelled probes hybridise to an amplicon and changes in the quenching of the fluorophore are detected. In collaboration with Zeneca Diagnostics Ltd and Oswel Research Products Ltd we are developing a new technology that is simple to use, gives highly specific information and avoids the major difficulties of the alternative methods. This uses a primer (which we will call a Scorpion primer) having an integral tail which is used to probe an extension product of the primer. The probing of a target sequence is thereby converted into a unimolecular event, which has substantial benefits in terms of kinetics, thermodynamics, assay design and probe reliability. We propose to develop Scorpion primers for use with a wide range of fluorophores and quenchers.
