Abstract

We have recently provided the first demonstrations that molecularly defined, heterologously expressed, Gi-protein-coupled receptors (GPCRs) can increase the phosphorylation state and cause activation of p70 S6 kinase. Agonists at these receptors are mitogenic and this is prevented by treatment of cells with a selective inhibitor of the p70 S6 kinase signalling cassette. We will now (1) establish the organisation and hierarchy of the protein components of this signalling cassette, (2) compare the regulation of this cassette by GPCRs and receptor tyrosine kinases, (3) define the contributions and points of bifurcation between GPCR-mediated activation of the p70 S6 kinase and ERK MAP kinase signalling cassettes, (4) examine how GPCR activation of the p70 S6 kinase cascade can regulate translational efficiency, and (5) determine the contribution of GPCR activation of the p70 S6 kinase signalling cassette to mitogenesis and regulation of the cell cycle.
