Abstract

The aim of this project is to elucidate the integration of T-cell antigen receptor- stimulated signalling cassettes which mediate thymocyte apoptosis. The project will be carried out using the novel technique of retroviral transfection of murine foetal thymic organ culture and will benefit from the experimental advantages provided by the dysfunctional apoptosis of the CD45-null thymus. Phase 1 will focus on the early events of TCR signal transduction coupling, in particular on Lck and Fyn. Phase 2 will investigate the Ras/phosphotyrosine phosphatase 1D and HS1 signalling cassettes and Phase 3 the integration of these signals with Bcl-2 family members. The project will show how the selection of highly specific signalling pathways regulates T-cell death and survival during thymic differentiation.
