Abstract

This project will study IL-4 driven cAMP/protein kinase A signalling in regulation of gene expression in human B lymphocytes. Preliminary work has demonstrated that IL-4-driven cAMP accumulation causes a loss of activity of protein(s) binding to a negative regulatory element (NRE) in the promoter of the IL-2 receptor alpha chain (CD25) gene. We will now characterise the NRE-binding protein (NRE-BP), define the mechanism by which it is regulated by the cAMP/PKA pathway and, in the later stages of the project, by other pathways. We will also explore the biochemical mechanism by which the IL-4 receptor is linked to cAMP generation, and will address the hypothesis that JAK family tyrosine kinases play a central role in this process.
