Abstract
The development of an efficient gene targeting system for the vascular endothelium is a pre- requisite for gene therapy to be fully exploited in diverse clinical situations. We have previously developed an endothelial cell-specific gene delivery system by incorporating novel targeting peptides and cell-specific promoters into adenoviral vectors. We wish to optimise gene delivery in vitro and in vivo and compare this for efficacy and persistence of gene expression to non-targeted adenoviral vectors. Using model therapies, the effect of overexpression of vasodilator genes from the endothelium in hypersensitive rats and the effect of overexpression of cytotoxin genes in the angiogenic endothelium of cancer will be evaluated.
