Abstract

The inheritable nature of DNA methylation patterns within the cell, coupled to the inhibitory effect of methylation on gene transcription, offers a novel 'single-shot' gene therapy approach, through the application of sequence targeted DNA cytosine methyltransferases. We have constructed a number of enzymes capable of specific methylation of predetermined sequences and we now propose to evaluate these enzymes for their in vivo ability to both deliver a methylation signal at the desired locus and repress transcription via a methylation mediated pathway. We also describe a number of experiments, where for the first time, the effects of variance in methylation density on transcription, chromatin structure and methylation spread, can be evaluated at a single defined site, within a constant model system.
