Abstract

The neuropeptide galanin is expressed at low levels in normal adult dorsal root ganglia (DRG) and up-regulates markedly following nerve injury. We have knocked out galanin demonstrating a novel survival and trophic role for galanin in the embryonic and adult DRG. The loss of a subset of small nociceptors almost completely abolishes the symptoms of neuropathic pain. This application will bring together cutting-edge molecular genetics and behavioural physiology, allowing us to identify novel genes that are the up- and down-stream components of the galanin-receptor interaction. These studies will further define the molecular cascades that mediate nerve regeneration and pathological nociceptive responses.
