Abstract

FRI causes plants to over-winter before flowering. FRI represses flowering by increasing RNA levels of floral repressor, FLC and this function is antagonised by vernalisation. Given the involvement of a Polycomb-type mechanism in vernalisation, we will test whether FRI functions like trithorax-group proteins to regulate FLC chromatin structure. We will use sector analysis to determine if FRI is required during a particular developmental window and chromatin immunoprecipitation to ask if FRI is associated with changes in the histone code at FLC. We will determine if FLC is a direct target of FRI and to identify proteins interacting with FRI throughY-2-H assays/plant functional analyses and affinity purification of complexes from plants.
