Abstract
We propose to demonstrate the potential of small molecular weight effectors based on aminoglycosides, coupled to RNA aptamers directed against cellular protein targets, for the dissection of complex metabolic pathways in vivo. As a test of this approach, we will use the transcription factors controlling amino acid biosynthesis in E. coli for methionine (MetJ), arginine (ArgR) and tryptophan (TrpR). The results will be assessed using gene arrays.
