Abstract
Although activation of E-cadherin adhesion initiates epithelial differentiation, the subsequent steps leading to polarised vectorial transport and intracellular sealing are less well understood. Trophectoderm differentiation in the mouse preimplantation embryo exhibits a protracted period (24 hours) between E-cadherin adhesion and epithelial function during which a temporally- controlled epithelial gene expression programme unfolds which we and our collaborator have characterised. We propose critical regulatory roles for tight junction ZO-1alpha+ is form and Na/K- ATPase Beta 1-subunit in completing differentiation. Molecular genetic, gene microinjection, immunoconfocal imaging and physiological approaches will be used to investigate our hypotheses.
