Abstract

In development there is much to learn of the genetic circuitry that operates downstream of patterning events to regulate cell differentiation. We will study this fundamental issue using a key transcription factor in Drosophila muscle development, Dmef2. We wish to test the idea that different muscle genes have distinct thresholds for activation by Dmef2 and then analyse the basis of these thresholds. We will identify candidate targets by microarrays using an allelic series expressing different levels of Dmef2. Our analysis of target gene enhancers will use computational methods including phylogenetically foot printing, and then we will experimentally dissect selected enhancers by mutagenesis of identified sites and in vivo expression assays.
