Abstract

Our understanding of the cascades that regulate the expression of pathway-specific activators of secondary metabolites is far from complete. Using biochemical approaches we have found, characterised and partially purified a factor that binds specifically to sites flanking the promoter of actII-ORF4. We now propose to (i) complete the identification of the specific DNA-binding factor and then (ii) create a knockout of the corresponding gene and study the phenotypic effects. In complementary experiments, we will also (iii) identify important nucleoside contacts within the binding sites and then determine the consequence of their mutation, comparing with results of (ii). Finally, we plan to (iv) identify other components in the regulation of actII-ORF4 and other possible targets for the specific DNA-binding factor.
